National Retinol Day 2023: History

Retinol was initially synthesized in 1947, after having been discovered in 1909 and isolated in 1931.

Frederick Gowland Hopkins was awarded the Nobel Prize in 1929 for his seminal work in 1912, which established the necessity of accessory elements undisclosed in milk for rodent growth. Elmer Verner McCollum, a biochemist associated with the University of Wisconsin, Madison, and his colleague Marguerite Davis identified a fat-soluble nutrient in butterfat and cod liver oil in 1913. The terminology used to refer to the “accessory components” was “fat-soluble A” in 1918 and “vitamin A” in 1920. In 1932, Swiss chemist Paul Karrer initially delineated the molecular structure of vitamin A.

Initially isolated in the 1930s, retinol was subsequently degraded by oxygen and sunlight. The unreliable substance led to the development of retinoic acid, which possessed greater stability. The initial investigations on retinoic acid documented its potential as an acne remedy in 1943. In the latter half of the 1950s, retinoic acid was introduced into the medical field for the treatment of acne.

Tretinoin, the most widely utilized retinoid, originated as a topical treatment for dermatological conditions in 1958. Its application in anti-aging therapies commenced during the 1980s. A 1983 publication suggested that retinoic acid could be used to treat aging of the epidermis. By the 1990s, retinoic acid had acquired a negative reputation for causing skin irritation and sensitivity; as a result, retinol, which was more skin-beneficial, supplanted it on the market.
