In 2019, Karolina placed second at the Prague Open and advanced to the quarterfinals of Wimbledon, where she defeated fellow Czech player Karoline Pliskova.

She was the first player since Li Na in 2006 to reach the Wimbledon quarterfinals in her début. In the same year, she reached the third round of the US Open but lost to Serena Williams. Karolina advanced to the semifinals of the 2021 Australian Open by defeating former world No. 1 Ashleigh Barty.
